Hepatitis E Virus (HEV) GENE DETECTION KIT

Intended Use

The kit is developed for the detection of viral RNA of the hepatitis E virus by reverse transcriptase/polymerase chain reaction (RT-PCR*).

Assay principle

Hepatitis E is an infectious viral disease with clinical and epidemiological features of acute hepatitis. Only seldom adult patients progress to fulminant hepatitis, but among pregnant patients the case fatality rate can be as high as 20%-30%. Hepatitis E is a waterborne disease transmitted primarily by contaminated water. There is also a possibility of zoonotic spread of the virus, since several non-human primates, pigs, cows, sheep, goats and rodents are susceptible to the infection. Hepatitis E virus (HEV) is a principal cause of acute hepatitis in adults throughout much of Asia, Middle East and Northern Africa and transmitted from person-to-person through the fecal-oral route. At least four major genotypes of HEV have been identified, only one serotype is recognized. HEV is endemic in developing countries where sanitary conditions are not well maintained.

After RNA extraction and reverse transcription viral RNA can be detected in stool and in serum of the infected patient by PCR*. For the detection of very low virus titers as it is regularly the case in serum samples it is required to perform a nested PCR*.

The HEV genotypes differ from each other; therefore, the kit contains two sets of reaction mixes. It is recommended to assay the samples with both sets of reaction mixes.

Contents:

The kit contains the lyophilised reaction mixes for running 20 specific RT reactions with primer A, 30 PCR* and 30 nested PCR* reactions with RxMix A, as well as 20 specific RT reactions with primer B, 30 PCR and 30 nested PCR* reactions with RxMix B. 

Required materials not provided:

  • Taq-Polymerase and Reverse Transcriptase
  • Agarose
  • TAE- or TBE electrophoresis buffer (can be included if required)
  • Sample buffer (can be included if required)
  • DNA-Standard (can be included if required)
  • Ethidiumbromide or other dyes for nucleic acid staining
  • Mineral oil for thermocyclers without heating lid
